I型胶原水凝胶的物质通透性评价

      Evaluation of Material Permeability of Type I Collagen Hydrogel

      • 摘要: 目的 为评价I型胶原蛋白水凝胶的物质通透性,建立体外物质通透性评价实验方法。方法 以BSA-FITC为指示物,组合PBS作为通透介质,借助转移小室(Transwell)建立体外物质通透性评价实验方法。结果 本研究建立的BSA-FITC定量检测方法在100 μg·mL-1~0.781 μg·mL-1的浓度范围内,其标准曲线的R2大于0.99,其定量下限(LLOQ)为3.125 μg·mL-1;RSD<5%;回收率在80%~120%范围内。结论 本研究建立了水凝胶类样品的物质通透性体外评价方法。利用该方法评价I型胶原蛋白水凝胶物质在28小时后其BSA-FITC通透率为100%。

         

        Abstract: Objective To establish an experimental method for evaluating material permeability of type I collagen hydrogels. Methods Using BSA-FITC as an indicator, by combining BSA-FITC with PBS they were used as permeability media, and using transwell load hydrogen sample to detect BSA-FITC transparent rate. Results In the concentration range of 100 μg·mL-1~0.781 μg·mL-1, the standard curve R2 ≥ 0.99, Lower Limit of Quantity (LLOQ) is 3.125 μg·mL-1, RSD <5%, detection recovery rate is in the range of 80%~120%. Conclusion In this study, we established an experimental method for evaluating material permeability of hydrogel. The BSA-FITC transparent rate of type I collagen hydrogel was 100% at 28 h.
