RhCE模型在医疗器械眼刺激评价中的应用潜力与挑战

      Progress in Application of RhCE Models in Eye Irritation Evaluation of Medical Devices

      • 摘要: 眼刺激评价是眼科医疗器械生物相容性评价的重要环节。随着实验动物“3R”原则的推进,3D重组人角膜上皮(reconstructed human cornea-like epithelium,RhCE)模型已成为替代传统Draize兔眼试验的研究热点。该文分析了传统动物试验的局限性,系统综述了RhCE模型的构建原理、生物学特性、主流商用模型应用现状,以及MTT组织活力、跨上皮电阻和炎症因子释放等评价指标的应用。探讨了RhCE模型在医疗器械监管中的采信现状、面临的挑战与发展前景,为我国医疗器械生物学评价的体外方法建设提供参考。

         

        Abstract: Eye irritation evaluation is a critical component of the biocompatibility assessment of ophthalmic medical devices. Driven by the advancement of the "3Rs" principles for experimental animals, 3D Reconstructed human Cornea-like Epithelium (RhCE) models have emerged as a prominent alternative to traditional Draize rabbit eye tests. This article analyzes the limitations of traditional animal testing and systematically reviews the construction principles, biological characteristics, and current application status of mainstream commercial RhCE models. Furthermore, the applications of evaluation metrics such as MTT tissue viability, transepithelial electrical resistance, and inflammatory cytokine release are discussed in detail. The current regulatory acceptance status, challenges, and future prospects of RhCE models in medical device regulation are explored, providing a reference for the development of in vitro biological evaluation methods for medical devices in China.
